== Function == Cholesterol needs to be transferred from the outer mitochondrial membrane to the inner membrane where cytochrome P450scc enzyme (CYP11A1) cleaves the cholesterol side chain, which is the first enzymatic step in all steroid synthesis. The aqueous phase between these two membranes cannot be crossed by the lipophilic cholesterol, unless certain proteins assist in this process. A number of proteins have historically been proposed to facilitate this transfer including: sterol carrier protein 2 (SCP2), steroidogenic activator polypeptide (SAP), peripheral benzodiazepine receptor (PBR or translocator protein, TSPO), and StAR. It is now clear that this process is primarily mediated by the action of StAR. The mechanism by which StAR causes cholesterol movement remains unclear as it appears to act from the outside of the mitochondria and its entry into the mitochondria ends its function. Various hypotheses have been advanced. Some involve StAR transferring cholesterol itself like a shuttle. While StAR may bind cholesterol itself, the exorbitant number of cholesterol molecules that the protein transfers would indicate that it would have to act as a cholesterol channel instead of a shuttle. Another notion is that it causes cholesterol to be kicked out of the outer membrane to the inner (cholesterol desorption). StAR may also promote the formation of contact sites between the outer and inner mitochondrial membranes to allow cholesterol influx.
==== Blood stains ==== There are several reddish stains on the shroud suggesting blood. McCrone (see painting hypothesis) showed that these contain iron oxide, and theorized that its presence was likely due to simple pigment materials used in medieval times. While the forensic doctor Pierluigi Baima Bollone initially claimed in 1983 to have identified type AB human blood along with traces of serum, aloes, and myrrh, this conclusion was later challenged by researchers like Alan Adler and more recently Kelly Kearse, who noted that early testing methods could not definitively confirm ancient human blood due to degradation and potential contamination. Skeptics cite forensic blood tests whose results dispute the authenticity of the Shroud, and point to the possibility that the blood could belong to a person who handled the shroud, and that the apparent blood flows on the shroud are unrealistically neat. As of 2025, it has not been scientifically demonstrated that the blood is of human, or even primate, origin.

== Complexity of the problem == After genomics and transcriptomics, proteomics is the next step in the study of biological systems. It is more complicated than genomics because an organism's genome is more or less constant, whereas proteomes differ from cell to cell and from time to time. Distinct genes are expressed in different cell types, which means that even the basic set of proteins produced in a cell must be identified. In the past this phenomenon was assessed by RNA analysis, which was found to lack correlation with protein content. It is now known that mRNA is not always translated into protein, and the amount of protein produced for a given amount of mRNA depends on the gene it is transcribed from and on the cell's physiological state. Proteomics confirms the presence of the protein and provides a direct measure of its quantity.

==== Magnetic Resonance Imaging (MRI) Detection ==== Over the past two decades, iron oxide nanoparticles for biomedical use had increased dramatically, largely due to its ability of non-invasive imaging, targeting and triggering drug release, or cancer therapy. Stem or immune cell could be marked with iron oxide nanoparticles to be detected by Magnetic resonance imaging (MRI). However, the concentration of iron oxide nanoparticles needs to be high enough to enable the significant detection by MRI. Due to the limited understanding of physicochemical nature of iron oxide nanoparticles in biological systems, more research is needed to ensure nanoparticles can be controlled under certain conditions for medical usage without posing harm to humans.
==== Early life stress ==== Epigenetic modifications including DNA methylation and histone acetylation regulate OPRK1 gene expression in response to environmental factors such as early life stress and psychological trauma. Decreased DNA methylation in intron 2 of OPRK1, functioning as a gene enhancer, has been observed in the anterior insula of individuals with histories of childhood abuse, which correlates with altered receptor expression and stress responsivity. Postmortem samples from suicide completers with a history of severe child abuse (CA) had higher rates of KOR downregulation relative to controls and suicide completers without CA history, an effect not accompanied by alterations in multiple other genes. Hypomethylation of OPRK1 intron 2 was associated with the CA group, as low levels of DNA methylation facilitate glucocorticoid binding and subsequent regulation of OPRK1 transcription.
